The ContiChrom Platform
The ContiChrom equipment platform consists of an integrated chromatographic equipment of hardware, software and documentation.
It is a continuous chromatography tool suitable for all purification modes with standard resin formats:
- ion-exchange
- mixed-mode (multi-modal)
- hydrophobic interaction
- affinity
- size exclusion
- reversed-phase
- hydroxyapatite
Capabilities
The principle of the ContiChrom platform is very flexible and
- is easily scalable from lab to pilot to production scale due to constant residence times and constant linear flow velocities
- can be run to fit different process challenges, e.g. 3-fraction to multi-fraction separations
- Example 3-fraction: ion-exchange purification of high titer microbially produced protein
- Example 3-fraction: reversed-phase purification of synthetically produced protein
- Example multi-fraction: simultaneous extraction of multiple plasma constituents such as hyperimmunes
Advantages
Operational cost savings: significant reduction due to
- faster process development due to reduced screening process
- higher productivities (up to 10x compared to batch chromatography) and lower buffer consumption (up to 70% less buffer)
- higher yields at required purity: up to 50%
Capital expenditure (CAPEX) savings:
- Match downstream throughput with upstream output: the ContiChrom platform allows the design of small biotech facilities with large production output. The large increase in upstream output due to high producer cell lines combines well with the high throughput capabilities of the ContiChrom equipment, resulting in low CAPEX.
- Reduced foot print: the high throughput capability of the MCSGP process results in large amounts of therapeutic proteins purified with small equipment sizes. Therefore, the DSP manufacturing foot print is significantly smaller.
