Enabling Purification Technologies - Ultra-high Resolution

Conventional batch chromatography suffers from three drawbacks:

  • there is a big discrepancy between the selectivity of analytical separations and preparative separations making it difficult to correlate analytical column chromatography profiles with preparative column chromatography profiles .
  • the preparative batch chromatography profile, being quite coarse, has to be fractionated and pooled fractions have to be further processed using an orthogonal separation matrix to obtain a reasonably pure product.
  • When scaling up for high throughput, batch chromatography created large back pressures that limit its scale and resolution

 

The ContiChrom platform can by-pass these disadvantages by employing the following principles that act synergistically to provide high resolution leading to high product yield and purity with superior throughput.

  • As several columns are used in continuous chromatography, the individual column dimensions are smaller, allowing using small particle matrices with high resolution power.
  • The lower back-pressure of the system allows to perform the run at higher pressures.
  • The internal re-cycling mode provides an enhanced engineering solution to a separation problem resulting in higher yield and purity.
  • The counter-current gradient elution mode enhances the resolution further.
