The purification of synthetic peptides is challenging, since the impurity profile is complex and overlaps with the desired product. This problem is accentuated for larger peptides, as the more synthesis cycles, the more potential impurities will accumulate and the yield will decrease.
Current preparative purification strategies using conventional HPLC chromatography suffer from low resolution, resulting in low product yield and/or purity as well as high solvent consumption. Re-chromatography of side fractions is used to increase yield but is often tedious and irreproducible, leading to poor process definition.
